Non-Contact Sub-500 nm IR and Simultaneous Raman Microscopy with Fluorescence Imaging Serves as a New Paradigm in Vibrational Spectroscopy

Optical photothermal infrared (O-PTIR) spectroscopy has established itself as a cutting edge vibrational microspectroscopy tool. It offers significant advantages over traditional FTIR, Quantum-Cascade Laser, and Raman spectroscopic tools, providing submicron simultaneous IR and Raman microscopy in a non-contact mode with high sensitivity. As a new modality, counter-propagating has been engineered to provide enhanced IR and Raman spatial resolution and sensitivity, when used with high numerical aperture refractive objectives.

This improves spatial resolution to approximately 300 nm for both IR and Raman, whilst also improving sensitivity, image quality, and facilitating immersion objective studies in fluid. Further, Kansiz’ team coupled a widefield epifluorescence to facilitate a novel concept, fluorescence co-located O-PTIR microspectroscopy. Several examples from life sciences, such as cells, both fixed and hydrated, bacteria and tissue will be provided to demonstrate these capabilities and how they can enable new experiments and new research findings.

Mustafa KansizMustafa Kansiz, Ph.D., is the director of product management and marketing for the mIRage IR microscope at Photothermal Spectroscopy Corp. with responsibilities in new product development, marketing, and applications development. He has over 25 years of experience working with FTIR, Raman microscopy, and imaging in routine and research applications in both industry and academia.

He has worked at Varian and Agilent Technologies serving in a range of technical and business development roles including FTIR microscopy and imaging product manager, product specialist, R&D scientist and European FTIR sales manager. He has a doctorate from Monash University in biotechnological application of FTIR spectroscopy and multivariate statistics.
